    Polycyclic aromatic hydrocarbons (PAHs) are known to have toxic effects on fish. In this study, we examined the effects of benz[a]anthracene (BaA), a type of PAH, on fish liver metabolism. Nibbler fish (Girella punctata) were intraperitoneally injected with BaA (10 ng/g body weight) four times over a 10-day period. BaA significantly decreased known bone metabolism-related plasma factors such as calcium and inorganic phosphorus. Moreover, significant reductions were observed in the plasma levels of known liver metabolism-related factors, including ferrous ions, total bile acids, total bilirubin, free bilirubin, aspartate aminotransferase, and alkaline phosphatase. Interestingly, mono-hydroxylated metabolites of BaA, such as 3 hydroxylbenz[a]anthracene (3-OHBaA), were detected in the bile of BaA-injected nibbler fish. This hydroxylated form of BaA was found in its free form, rather than conjugated with glucuronic acid or sulfuric acid. Due to the lack of whole-genome sequence data for the nibbler fish, two nibbler fish-specific apoptosis-related factors (TNF receptor superfamily member 1A: tnfrsf1a and TNF superfamily member 10: tnfsf10) were isolated by De novo RNA sequencing. In a liver tissue culture, 3-OHBaA (10−6 M) significantly upregulated the expression of tnfrsf1a and tnfsf10 in the liver. These results provide the first evidence that 3-OHBaA metabolites exhibit toxic effects on the liver in teleost.

    We have cloned and characterized a novel gene from both human and mouse that encodes a new member of the immunoglobulin superfamily. The gene is preferentially expressed in both brain and testis, and hence, termed BT-IgSF (brain- and testis-specific immunoglobulin superfamily). The predicted protein consists of V-type and C2-type immunoglobulin domains as well as a hydrophobic signal sequence, a single transmembrane region, and a cytoplasmic domain. Human BT-IgSF protein (431 amino acids) is 88% identical to the mouse protein (428 amino acids) and both show significant homology to coxsackie and adenovirus receptor (CAR) and endothelial cell-selective adhesion molecule (ESAM). We examined the expression of BT-IgSF with various cultured cells and found that the gene was expressed in both neurons and glial cells in vitro. Furthermore, the expression was preferentially detected in pyramidal cell layers of the dentate gyrus and hippocampus and in commissure fibers of the corpus callosum, in brain tissue sections examined.     We identified two phosphorylatable proteins (33 kDa and 48 kDa) in crude extractes of spermatozoa from the sea urchin, Hemicentrotus pulcherrimus. While the 33 kDa protein was phosphorylated in 100 mM Mg^<2+>-containing medium with cAMP or cGMP, phosphorylation of the 48 kDa protein occurred irrespective of the Mg^<2+>-concentration if it was more than 10 mM. Physiological concentration if it was more than 10 mM. Physiological concentrations of cAMP or cGMP triggered dephosphorylation of the [^<32>P]-phosphorylated 48 kDa protein in a concentration dependent manner. The dephosphorylation of 48 kDa protein was inhibited with calyculin A or okadaic acid. The 48 kDa protein was associated with a 39 kDa protein to form a larger oligomer with about 400 kDa. The purified 400 kDa protein showed cyclic nucleotide-dependent histone kinase activity. The histone kinase activity shifted from 400 kDa to 39 kDa on a Superose 6 HR in the presence of 5×10^<-6>M cAMP. Phosphorylation of the 48 kDa protein in the purified 400 kDa protein required only Mg[^<32>P]ATP. A photoaffinity cAMP analogue, 8-N_3-[^<32>P]cAMP, was incorporated into the 48 kDa protein. These results suggest that the 48 kDa protein which is a reguratory subunit of the histone kinase is autophosphorylated, and binding of cAMP to the phosphorylated 48 kDa subunit of the kinase results in dissociation of the 48 kDa subunit(s) from the 39 kDa subunit(s) to allow the 48 kDa subunit accessible to a protein phosphatase.
