ケラチン遺伝子変異の違いによる単純型表皮水疱症の病態への影響と個別化医療の開発
基盤研究(C)
中間径フィラメントであるケラチン（keratin: K）は、細胞骨格として組織の構造と強度を維持する。単純型表皮水疱症では表皮基底層に発現するK5/K14の遺伝子変異により機械的水疱を生じる。なかでもK14遺伝子の新規変異を有する家系は強い炎症や痒みを伴ったことから、細胞骨格の異常が炎症反応などに影響するとの着想に至った。先行論文の知見から、K14変異がミトコンドリアの分布・機能・代謝の異常を起こし、自然免疫異常や創傷治癒遅延を誘導するとの仮説を立てた。ドキシサイクリン誘導性に変異K14を発現する不死化表皮細胞株を作製し、①K14変異の違いによるミトコンドリア機能およびオートファジーの差を解析するとともに、②炎症反応や創傷治癒に与える影響を明らかにする。本研究により、K線維の未知の細胞生物学的機能を明らかにし、新規治療の開発や既存治療の応用、変異の差異に基づく個別化医療の確立、他のK線維異常疾患の病態解明への貢献を目指す。
